The PLoS Pathogens study reports on nce‑miR‑12220, a microRNA identified in Nosema ceranae spores, and its interactions with honeybee (Apis mellifera) midgut epithelial cells. The authors combined target prediction, dual‑luciferase assays and fluorescence in situ hybridization with RNA interference and gain‑ and loss‑of‑function experiments. They report sequence‑specific interaction of nce‑miR‑12220 with ATP‑A and γ‑tubulin binding regions and detection of the miRNA in infected midgut epithelial cells. Manipulating the miRNA altered transcript levels of those targets: overexpression increased ATP‑A and γ‑tubulin transcripts and inhibition produced the opposite change.

Experimental readouts described in the same paper linked nce‑miR‑12220 manipulation to host and parasite phenotypes. Silencing either ATP‑A or γ‑tubulin reduced expression of the parasite virulence‑associated gene NcRBL and improved worker survival relative to a scramble control. Overexpression of nce‑miR‑12220 was reported to reduce expression of Toll‑pathway‑associated genes (Cactus and dorsal) and antimicrobial peptide genes (Defensin and Hymenoptaecin), to increase N. ceranae spore load and sucrose consumption, and to decrease midgut ATP content; inhibition reversed those responses and reduced parasite burden. The authors note that survival changes after miRNA manipulation trended as predicted but did not reach statistical significance.

The study frames nce‑miR‑12220 as a parasite‑derived regulator that promotes N. ceranae proliferation while reshaping host immune and energetic responses, and suggests the miRNA and its target network as candidates for controlling bee nosemosis. As reported, the findings are based on molecular and experimental manipulations in infected workers; the reported survival result did not achieve statistical significance, and further work would be required to assess broader applicability and practical control strategies.
